Summary

An increased rate of intramammary infections due to environmental and opportunistic pathogens (coagulase negative staphylococci) has been observed in recent years. The presence of these microorganisms is more closely linked to the hygiene of the environment and/or the lower activity of mammary gland defences. Within a research project aimed at defining the immune status of ewesâ mammary gland and its influence on mastitis development, this paper reports the results of the assessment of phagocytic activity of ewes and its relationship with intramammary infections. A complete lactation follow-up study was conducted on the PMNs of 40 ewes from a Sardinian commercial herd. Milk samples (100ml) were collected before the morning milking every 4 weeks. Phagocytic activity was estimated by luminol enhanced chemiluminescence. The area under the curve values was weighed by the number of viable PMNs. Overall, 79 samples resulted in being infected by CNS or environmental streptococci. Healthy udders were shown to have a significantly lower PMNs rate (36% in comparison with 72% observed in infected udders) and a higher rate of macrophages (49% vs. 20%). However, data showed that phagocytic activity was significantly higher in bacteriologically negative udders (128 mV/1000PMNs) in comparison with the values observed in infected udders (10 mv/1000 PMNs). These results confirm that intramammary infection is associated with a decrease in the immune status of the mammary gland.
