The present research aimed to use molecular markers to study rapid detection of self-biting disease. We used RAPD marker to assess the level of genetic differentiation between self-biting and healthy blue fox group. A single RAPD marker S414 amplified a specific band of 768bp in self-biting of blue fox, which was designated as SRS414. The sequences of the bands exhibited 70% similarity to Macaca mulatta MHC class I region. The RAPD marker was converted into SCAR marker via cloning and sequencing of RAPD amplification and designing of 18-mer oligonuleotide primers. The RAPD and SCAR marker were validated in two groups respectively. χ² test showed significant difference (P<0.05) between the detection rate of the two groups. This indicated that SCAR-S414 can be used as a positive marker to detect self-biting in farm. Furthermore, the finding that self-biting linked with MHC genes has implications for the mechanism of this disease.
