Although methods for the effective treatment of wastewater exist and are essential to ensure the safety of drinking water and crops, the wastewater originated from dairy and cattle recovery may harbour bacterial species including human pathogens such as Escherichia coli O157:H7. Enterohaemorrhagic Escherichia coli O157:H7 is one of the most studied foodborne pathogen bacteria, because of its widespread diffusion, low dose infectiveness and severe symptoms associated. Since dairy and cattle are considered a major reservoir of Escherichia coli O157:H7 this pathogen may potentially infect the drinking water supply from cattle waste wash water. Conventional culture techniques for the detection of E. coli O157:H7 in water and environment are labour intensive and time consuming and often fail the recovery of this bacteria when the concentration in the sample is low (beyond the threshold of sensitivity) or when cells enter in viable not culturable (VNC) state. In this study we have developed and evaluated a molecular method based on real-time PCR for the identification of E. coli O157:H7 in cattle and dairy wastewater samples produced from Mozzarella cheese factories, without pre-enrichment step before DNA extraction. Moreover, we were able to identify E. coli O157:H7 in cattle and dairy wastewater in which previous method (including standard PCR) failed to work.
