Antibody-based immunoaffinity columns clean-up combined with HPLC/ fluorescence detection has become very common in mycotoxin analysis of foodstuffs, feedstuffs and biological fluids due to a large number of advantages over other commonly used clean-up procedures. Immunoaffinity columns are commercially available for aflatoxins B₁, B₂, G₁, G₂ and M₁, ochratoxin A, fumonisins B₁, B₂, and B₃, zearalenone, deoxynivalenol, and T-2 and HT-2 toxins. Recently, commercial multimycotoxin immunoaffinity columns have been used for simultaneous determination of T-2 and HT-2 toxins, aflatoxins B₁ and M₁, and ochratoxin A, or aflatoxins B₁, B₂, G₁, and G₂, ochratoxin A, and zearalenone by HPLC/fluorescence detection with good accuracy and precision. Several methods using HPLC with fluorescence detection and antibody-based immunoaffinity columns clean-up have been validated by collaborative studies for various mycotoxins in a number of food commodities, and have been adopted as official or standard methods by AOAC International or the European Committee for Standardization.
