Wood hedgehog (Hydnum repandum) is an edible ectomycorrhizal (EMC) mushroom found in conifers and broad-leaved trees forests, and close to chanterelle (e.g. Cantharellus cibarius) mushroom. EMC fungi are essential in maintaining forest health and resilience, each EMC fungus species having a different environmental impact. Biochemical and physiological properties of various species should be thoroughly investigated in order to better understand their influence on ecosystems. In this research, the activity of polyphenol oxidase (PPO), enzyme responsible for browning reactions, was investigated and characterized in H. repandum. At least one soluble PPO isoenzyme (~120 kD) that oxidized monophenols and diphenols, was detectable in H. repandum extract. Maximum limiting reaction rate and highest catalytic efficiency were found with p-cresol as substrate. Activation by SDS and inhibition by kojic acid were observed to various extents depending on the substrate. Data were compared with those previously obtained with C. cibarius, a close species in the same habitat.
