We report the cloning and characterization of 21 new metallothioneins (MTs) from five Tetrahymena species: T. patula, T. borealis, T. elliotti, T. americanis and T. malaccensis. 12 of them are putative CdMTs (subfamily 7a) and 9 have been identified as CuMTs (subfamily 7b). We have also isolated complete cDNAs of some of these new sequences. Moreover, we report one CuMT from the fish ectoparasite ciliate Ichthyophthirius multifiliis. The comparative in silico analysis of these 22 new MTs with those 18 previously described in different species of Tetrahymena corroborates the division of the family 7 (ciliate MTs) into two subfamilies (7a and 7b). The main differences between them are: (a) a unique Cys pattern; (b) a different relation between Lys and Cys residues; (c) the strict modular structure of CdMTs; and (d) the asymmetry of the codon usage for Gln residues. A comparative study of all ciliate MTs with those from other organisms shows the presence of unique features with regard to standard MTs (mainly from vertebrates).
