This work was aimed to evaluate the effect of Pseudomonas aeruginosa UCP/WFCC 0099 wild-type and acclimated on diesel in the oxidation of diesel fuel and sunflower biodiesel. A factorial 2×2 design was used for each these cultures and for the wild-type strain. Also was varied the substrate using diesel or sunflower biodiesel, being in total six factorial designs. The fuels degradation tests were carried out using Erlenmeyer´s flasks containing 100 ml of Bushnell-Hass medium added of 0.5 μg of the indicator 2,6-dichlorophenol indophenol (DCPIP). Different concentrations of microbial inoculums to each substrate established for factorial designs with 12 test and 4 repetitions of central point were used. The microbial potential of fuels biodegradation was carried out by visualization of a decolourization of the medium, followed optical density at 610 nm determinations. The best results showed degradation of 78.01%, 93.93% and 95.85% for diesel, and for sunflower biodiesel was verified 98.03%, 98.92% and 99.87% of degradation for the wild-type strain, the strain acclimatized to 15 and 30% of diesel, respectively.
