Species belonging to Aspergillus of sections Circumdati and Nigri can produce ochratoxin A (OTA) and their occurrence in Spanish barley indicates an adaption to warmer weather. The aims of this work were to identify the most relevant OTA producing Aspergillus spp. by PCR and to study the effect of environmental conditions (a_w, temperature cycle) on the development of these fungi and on OTA production in Spanish barley. The most important OTA-producing Aspergillus spp. were isolated and identified by species-specific PCR protocols based on the ITS1 and ITS2 regions. Working under different conditions growth was assessed and OTA level in cultures was determined by solvent extraction, immunoaffinity column clean-up and LC-fluorescence detection. Aspergillus steynii and Aspergillus carbonarius are good indicators of OTA risk in barley. Temperature, a_w, and isolate influenced OTA production. The highest OTA levels were found in barley cultures of A. steynii at 0.98 aw and at 22/32 °C cycle.
