In the present study bacterial isolate C27 from soil sample was tested for chitinase production on solid medium. It was transferred to liquid medium and further culture conditions were optimized. The influences of different carbon and nitrogen sources on enzyme production were investigated. It was established that colloidal chitin, used as a primary carbon source, induced the highest enzyme activity – 1.62 IU/ml at 48^th h of cultivation. Necessity of second carbon source was satisfied by starch, which shown activity 3.78 IU/ml. As nitrogen source best results were shown by yeast extract with 3.29 IU/ml. The optimization of nutrient levels was done using respond surface design methodology. The model showed the significant influence of colloidal chitin on chitinase production by isolate C27/5. Inoculums age and volume, temperature and primary pH of the medium were also investigated.
