Ochratoxin A (OTA) is a fungal metabolite produced by some Aspergillus and Penicillium genera that are found in a variety of plant food products such as cereals. OTA has been shown to affect proliferation and migration of neurons in the developing rodent brain and may also affect to neurogenesis. In that sense, previous studies in vitro have tested the vulnerability of hippocampal neural stem cells (NSCs) of adult mouse to the toxin. The subventricular zone (SVZ) of mammals is other of the big neurogenic niche of the brain. For that reason, in the present work, we have studied how OTA exposure can affect to survival, proliferation and differentiation of NSC cultured from the SVZ. The results of the viability after 24, 48 and 72 h showed that the percentages of attached cells were decreased in a dose- and time-dependent manner under OTA exposure in both proliferation and differentiation media. In addition, NSCs were not able to differentiate into astrocytes and young neurons when OTA was over 1 μg/ml. By decreasing cell proliferation and differentiation, OTA can be limiting the developing process of the brain in embryos and the regenerative capacity of the SVZ in the adults and, therefore, affecting to the natural mechanism of brain repair.
