Chloramphenicol (CAP), a ‘broad-spectrum’ antibiotic known to bind 50S prokaryotic ribosomes thus preventing translation, has been identified as a source of numerous disorders including mitochondrial protein synthesis alteration, cancer progression, cancer cell invasion, leukemiogenesis, and fatal aplastic anemia. CAP’s toxicity has been in part attributed to its potential role in apoptosis for which it is sometimes reported as an inhibitor, sometimes as an inducer, depending on the system being studied. In this research, the effect of CAP on the yeast Candida utilis was investigated by morphological studies conducted by thin section and freeze-fracture electron microscopy and by concomitant studies on cytochromes biosynthesis, respiration, and cell growth and yield. CAP inhibited mitochondrial cytochromes biosynthesis, was an uncoupler of oxidative phosphorylation, and produced apoptosis and necrosis in this strain of yeast. Morphometric analysis showed that CAP-grown cells were up to 30% larger in volume than control cells and occasionally contained giant peroxisomes.
