This study investigated high voltage ACP efficacy for inactivation of monoculture biofilms of Staphylococcus aureus, Listeria monocytogenes, Escherichia coli and Pseudomonas aeruginosa. Biofilms grown for 48 h in 96 well microtiter plates (10^6-7cfu/ml), were sealed within a high barrier polypropylene bag before atmospheric cold plasma (ACP) treatment. Direct or indirect ACP discharge was generated in atmospheric air using 80 kV_RMS for 0-300 s. Following ACP treatment, samples were left unopened at room temperature for 24 h prior to microbiological analysis. The effect of ACP treatment was quantified by colony count and XTT assay. Reduction of biofilm was substantial but dependent on the type of bacterial strain. S. aureus biofilms were reduced by 5 and 4.3 log₁₀ cfu/ml, and a 97% and 94% reduction in metabolic activity was recorded using the XTT assay after direct and indirect ACP treatment for 300 s, respectively. L. monocytogenes viable cells were reduced by 5.2 and >5.3 log₁₀ cfu/ml after direct and indirect ACP treatment for 300 s, respectively (91% and 96% by XTT assay). No colonies were detected for gram negative E. coli and P. aeruginosa after 60 s using either mode of exposure. However, for P. aeruginosa biofilms, following extended ACP treatment, approximately 40% of cells were viable but not culturable (VBNC) as determined by retention of metabolic activity. The potential application of high voltage in-package ACP treatment for reduction of bacterial biofilms has been demonstrated. Rapid decontamination times are useful for treatment of heat sensitive materials. However, further studies are required to characterise the effect of extended and pulsed ACP treatment regimes for biofilm inactivation and removal to determine conditions eliminating the possibility of VBNC state of bacteria.
