Consumption of maize contaminated with Fusarium sp and FB₁ has been associated with human esophageal cancer (EC) in the Transkei, South Africa. The southern Brazilian states of Santa Catarina, Parana and Rio Grande do Sul also have a high incidence of EC. Fumonisin B₁ (FB₁) a metabolite of the common maize contaminant F. verticilioides induces liver and kidney cancers in experimental mice and rats. However, the specific role of the fumonisins in the etiology of human disease, including EC, remains to be determined. Recent studies using maize from the Transkei have shown the presence of a putative genotoxin. Maize for human consumption from SC state, previously shown to be predominantly infected with F. verticillioides was analysed for the presence of the putative genotoxin by incubation of a methanolic extract with DNA in the presence of rat liver S9. Of the 76 samples, only maize samples from the western region of the state produced the Fusarium-specific DNA adduct. A similar adduct was present from extracts of maize infected by F. verticillioides MRC826 and RRC415 from South Africa and Mississippi, respectively. These results suggest the Fusarium mycotoxin responsible for the DNA adduct is not widespread in the samples collected from Santa Catarina in the fall of 2000.
