Barley is rich in important dietary fibre components, such as β-glucans and arabinoxylans. In this work the fermentation of fibre fractions isolated from barley by faeces from human infants was investigated. Four fibre fractions were prepared: Ground barley flour (GCM); purified water soluble material after amylase treatment containing 82% β-glucan (WSM-AU); water insoluble material (WUM); and the remaining water insoluble material after alkali treatment of WUM, including insoluble arabinoxylans and β-glucans (WUM-B-RES). Fermentation rate, production of short chain fatty acids (SCFA) and changes in the microbiota was compared. Results from in vitro fermentation showed that the fibre fraction WSM-AU, gave a 71% reduction in polymeric glucose, a high gas production and an evident pH reduction during fermentation. Similar results were obtained for barley flour (GCM), but GCM had a higher reduction in polymeric glucose (77%). The water insoluble fibre fraction, WUM, was fermented at the lowest rate of all barley fibre fractions tested, giving a low gas production, a low pH reduction and a 28% reduction in polymeric glucose. The treatment of the water insoluble fibre fraction, WUM, with alkali, made it possible for faeces bacteria to ferment xylose and arabinose, 46% and 36% respectively in the resulting WUM-B-RES. This fraction showed a lower gas and pH reduction as well as a lower reduction in polymeric glucose compared to WSM-AU and GCM. The barley fractions differed less in short chain fatty acid (SCFA) formation. However, the β-glucan fraction WSM-AU gave the highest level of SCFA, mostly acetic acid and propionic acid. WSM-AU significantly increased taxonomic group Bacteroides while reduced Coriobacterineae. WUM and WUM-B-RES, gave a significant increase in unclassified bacteria as well as a decrease in taxonomic group Coriobacterineae. Finally, the ground barley flour, GCM, showed only a small, but significant increase in taxonomic group Lachnospira. In conclusion, different rates of in vitro fermentation between barley fractions indicated that water soluble and insoluble non-starch polysaccharides may function differently as substrate for bacteria in the gastric intestinal tract of juveniles.
