Water soluble modified polysaccharides have been used in food as performance additives for many decades. In recent years there has been increased awareness that some modified polysaccharides resist digestion and fermentation in the human gastrointestinal tract. Any modified polysaccharides which also prove to provide beneficial health properties such as reducing hypercholesterolemia or preventing diabetes could become an important source of dietary fibre. Analytical methods to determine their content in food are required. Dietary fibre is most often determined by well established enzymic-gravimetric methods which simulate gastro-intestinal digestion and then rely on ethanol to quantitatively precipitate the soluble fibres. Some modified polysaccharides survive the digestion process intact but will not precipitate under conditions employed by existing methods. This paper highlights size exclusion liquid chromatography as a technique for the determination of select modified polysaccharides which are not accurately determined by existing AOAC methods. Results are provided illustrating the application of size exclusion chromatography to the determination of 0.2-27% methyl cellulose and hydroxypropyl methylcellulose in food. The method provides acceptable precision and recovery ‘on par’ with existing dietary fibre methods. This method and chromatographic technique could be extended to analysis of other modified polysaccharides for which no suitable method exists.
