One of the basic backbone units of pectin is polygalacturonic acid (PGA), a homopolymer of α-1,4-linked galacturonic acid (GalA). PGA synthase (PGA α-1,4- galacturonosyltransferase) catalyzes the successive transfer of GalA onto the nonreducing end of oligogalacturonic acid to synthesize PGA. PGA is thought to consist of consecutive GalA residues with the degree of polymerization (DP) around 100 in native molecules. However, it has not been experimentally proven that PGA synthase enables elongation to such a length of PGA chain. In vitro synthesis of PGA has been partially successful with production of pyridylaminated oligogalacturonic acids (PAOGAs) with DP of up to 27. In this article, we describe experimental conditions for generating longer PGA in vitro. A microsomal fraction prepared from azuki bean epicotyls was solubilized with CHAPS (20 mM) in a buffer and the solubilized enzyme reacted with PA-OGA of DP 12 and UDP-GalA in a buffer containing 10 mM potassium oxalate. The enzyme was able to transfer more than 30 GalA residues and elongated the chains up to PA-OGA of DP 45.
