Our knowledge of how the structural and biochemical complexities of pectic polysaccharides are integrated with cell wall microstructures and with cell and tissue biology is limited. Monoclonal antibodies to defined structural elements of pectic polysaccharides are essential tools to map pectic structures in relation to cellular and developmental processes and in relation to taxonomic diversity as a prelude to the elucidation of functions. Restricted patterns of cell wall epitope occurrence within individual cell walls or across sections of plant materials are well documented and absence of an antibody binding has generally been taken as absence of the polymer. Two emerging strands of work indicate that patterns of cell wall epitope occurrence must be interpreted with caution. Firstly, recent work has indicated that cell wall hemicellulose epitopes can be effectively masked by pectin. Secondly, a new monoclonal antibody to 1,5-arabinan, LM13, has binding properties distinct from the previously isolated and established anti-1,5-arabinan monoclonal antibody LM6. This pair of antibodies indicates that the structure of 1,5-arabinan in muro is highly regulated and specific cell-related occurrences of the epitopes can vary between organs and between species.
